基于新一代測序技術的A2O與BIOLAK活性污泥宏基因組比較分析

本研究是(shi)(shi)A2O(anaerobic/anoxic/oxic,厭氧(yang)(yang)/缺氧(yang)(yang)/好(hao)氧(yang)(yang))與百(bai)(bai)(bai)樂(le)克(ke)(BIOLAK)活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)宏(hong)(hong)(hong)基(ji)(ji)(ji)因(yin)(yin)(yin)(yin)組(zu)(zu)(zu)(zu)比(bi)(bi)較分(fen)析的(de)(de)首份報告.在(zai)(zai)(zai)百(bai)(bai)(bai)樂(le)克(ke)和(he)(he)A2O活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)宏(hong)(hong)(hong)基(ji)(ji)(ji)因(yin)(yin)(yin)(yin)組(zu)(zu)(zu)(zu)中(zhong)(zhong),分(fen)別檢(jian)測(ce)到47個(ge)和(he)(he)51個(ge)門(men)類,超過了澳大利亞(ya)強化生(sheng)物(wu)(wu)除(chu)磷(lin)(enhancedbiologicalphosphorusremoval,EBPR)、美國強化生(sheng)物(wu)(wu)除(chu)磷(lin)和(he)(he)Bibby活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)中(zhong)(zhong)所檢(jian)測(ce)到的(de)(de)生(sheng)物(wu)(wu)類群(qun).百(bai)(bai)(bai)樂(le)克(ke)活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)中(zhong)(zhong)所發現的(de)(de)門(men)類,均(jun)在(zai)(zai)(zai)A2O活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)中(zhong)(zhong)檢(jian)測(ce)到.但是(shi)(shi),有(you)(you)4個(ge)門(men)類僅在(zai)(zai)(zai)A2O活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)中(zhong)(zhong)出(chu)現.Ignavibacteriae門(men)在(zai)(zai)(zai)A2O活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)宏(hong)(hong)(hong)基(ji)(ji)(ji)因(yin)(yin)(yin)(yin)組(zu)(zu)(zu)(zu)中(zhong)(zhong)的(de)(de)比(bi)(bi)例(li)(li)(2.0440%)是(shi)(shi)百(bai)(bai)(bai)樂(le)克(ke)活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)(0.6376%)的(de)(de)3.2倍(bei).與此(ci)同(tong)時,芽單胞菌門(men)在(zai)(zai)(zai)百(bai)(bai)(bai)樂(le)克(ke)活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)宏(hong)(hong)(hong)基(ji)(ji)(ji)因(yin)(yin)(yin)(yin)組(zu)(zu)(zu)(zu)中(zhong)(zhong)的(de)(de)比(bi)(bi)例(li)(li)是(shi)(shi)2.4673%,比(bi)(bi)其在(zai)(zai)(zai)A2O活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)中(zhong)(zhong)的(de)(de)比(bi)(bi)例(li)(li)(0.1404%)高(gao)出(chu)17倍(bei)多(duo);衣原體門(men)在(zai)(zai)(zai)百(bai)(bai)(bai)樂(le)克(ke)活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)宏(hong)(hong)(hong)基(ji)(ji)(ji)因(yin)(yin)(yin)(yin)組(zu)(zu)(zu)(zu)中(zhong)(zhong)的(de)(de)比(bi)(bi)例(li)(li)是(shi)(shi)0.2192%,比(bi)(bi)其在(zai)(zai)(zai)A2O活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)中(zhong)(zhong)的(de)(de)比(bi)(bi)例(li)(li)(0.0360%)高(gao)出(chu)6倍(bei)多(duo).另外,在(zai)(zai)(zai)屬的(de)(de)層(ceng)級,有(you)(you)167個(ge)屬僅在(zai)(zai)(zai)A2O活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)中(zhong)(zhong)檢(jian)出(chu);與此(ci)同(tong)時,有(you)(you)50個(ge)屬僅發現于百(bai)(bai)(bai)樂(le)克(ke)活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)中(zhong)(zhong).因(yin)(yin)(yin)(yin)此(ci),在(zai)(zai)(zai)門(men)和(he)(he)屬的(de)(de)層(ceng)級,A2O與百(bai)(bai)(bai)樂(le)克(ke)活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)的(de)(de)生(sheng)物(wu)(wu)群(qun)落均(jun)存(cun)在(zai)(zai)(zai)巨大差異.然而,在(zai)(zai)(zai)百(bai)(bai)(bai)樂(le)克(ke)和(he)(he)A2O兩個(ge)活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)中(zhong)(zhong),與氮(dan)(dan)、磷(lin)、硫(liu)和(he)(he)芳香族化合物(wu)(wu)代謝相(xiang)(xiang)關的(de)(de)功能(neng)基(ji)(ji)(ji)因(yin)(yin)(yin)(yin)比(bi)(bi)例(li)(li)均(jun)是(shi)(shi)非常接近的(de)(de).此(ci)外,兩個(ge)活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)中(zhong)(zhong)KEGG(KyotoEncyclopediaforGenesandGenomes,京(jing)都基(ji)(ji)(ji)因(yin)(yin)(yin)(yin)與基(ji)(ji)(ji)因(yin)(yin)(yin)(yin)組(zu)(zu)(zu)(zu)百(bai)(bai)(bai)科全(quan)書(shu))所有(you)(you)6個(ge)類別的(de)(de)排(pai)序均(jun)相(xiang)(xiang)同(tong).同(tong)時,氮(dan)(dan)代謝相(xiang)(xiang)關的(de)(de)功能(neng)基(ji)(ji)(ji)因(yin)(yin)(yin)(yin)分(fen)類和(he)(he)通路分(fen)析表明,高(gao)豐度酶在(zai)(zai)(zai)百(bai)(bai)(bai)樂(le)克(ke)和(he)(he)A2O宏(hong)(hong)(hong)基(ji)(ji)(ji)因(yin)(yin)(yin)(yin)組(zu)(zu)(zu)(zu)中(zhong)(zhong)具有(you)(you)相(xiang)(xiang)同(tong)的(de)(de)排(pai)序.因(yin)(yin)(yin)(yin)此(ci),A2O和(he)(he)百(bai)(bai)(bai)樂(le)克(ke)活(huo)(huo)(huo)性(xing)(xing)(xing)污(wu)(wu)(wu)(wu)泥(ni)(ni)宏(hong)(hong)(hong)基(ji)(ji)(ji)因(yin)(yin)(yin)(yin)組(zu)(zu)(zu)(zu)比(bi)(bi)較分(fen)析顯示(shi),兩個(ge)不(bu)同(tong)的(de)(de)生(sheng)物(wu)(wu)群(qun)落具有(you)(you)相(xiang)(xiang)似(si)的(de)(de)功能(neng)分(fen)配.
